How to Use

Silver Staining

Fix, wash, and then incubate the gel in Sensitizer Solution. Wash and incubate the gel in Stain Solution. Wash and develop the gel in Developer Solution. Stop the reaction when desired band intensity is achieved with Stop Solution.

Silver Destaining

Submerge the gel in a solution comprised of Solution A & B.  Gently agitate the gel until the desired destaining has been achieved.  Stop the reaction by rinsing the gel with deionized water.

Silver Staining - Destaining Kit

The Silver Staining-Destaining Kit is a combination of two products for convenience; Silver Bullit™ Silver Stain Kit and Silver Subtract™ Silver Destaining Reagent. Proteins can be detected in polyacrylamide gels with high sensitivity and nearly undetectable background using AMRESCO's Silver Bullit™ Silver Stain kit. The kit's colorimeteric staining procedure allows detection of subnanogram levels of protein, which is 100-fold more sensitive than Coomassie® Blue staining.

Gels stained with Silver Bullit® or any other silver staining product can be completely destained with Silver Subtract™ Silver Destaining Reagent. It is an excellent kit for the removal of silver ions before re-staining with silver stain. Silver Subtract™ is also compatible with subsequent visualization of the gel using fluorescent, chemiluminescent, or chromogenic stains, such as Coomassie® Blue. Partial destaining can also be achieved with more dilute reagents to remove high background, reduce staining of overloaded gels, or remove artifacts and even background.

Mid/Low Range Protein Molecular Weight Marker (J450) was resolved on a 15 % NEXT GEL® (M258) and subsequently stained with the Silver Staining - Destaining Kit using the 30 minute fast protocol.
