Hyptonic Lysis Buffer
AMRESCO’s Hypotonic Lysis Buffer is a detergent-free, ready-to-use solution for the isolation of cytoplasmic proteins from cultured mammalian cells. Cell lysis and subsequent isolation of the crude cytoplasmic fraction by a simple centrifugation step is complete in less than 30 minutes. The protocol is compatible with a variety of downstream applications including Western Blotting and ELISA procedures as well as protein assays which are sensitive to the presence of detergents in the lysis buffer.

Cellular lysis determinded by protein electrophoresis and western blot using detergent-based buffer or hypotonic lysis buffer
Proteins isolated from K562 cells using AMRESCO’s Cytoplasmic/Nuclear Protein Enrichment Kit (
M330, lane A1, B1) and AMRESCO’s Hypotonic Lysis Buffer (M334, lane A2, B2) were run on a 12.5% Fluorescent SPRINT NEXT GEL® (
M318) for 30 minutes at 300V. Proteins were visualized by UV transillumination (Figure A), then transferred to PVDF membrane and immunoblotted with β-actin antibody using the Rapid Western Blotting Kit-Rabbit-SDT (
N791). β-actin was detected using VisiGlo Plus™ HRP Chemiluminescent Substrate Kit (
N219, Figure B).
