EZ-Vision® In-Gel Sensitivity

DNA (2.5 ng - 80 ng)was mixed with 6X Agarose Gel Loading Buffer and applied to a 1% TAE agarose gel. Samples were resolved for 1 hour at 6.4 V/cm in 1X TAE buffer. Fluorescent visualization of DNA was achieved with a 500 msec UV exposure using a Sybr Green filter with the Syngene GBox-HR Gel Doc System.

RNA EZ-Vision vs. Ethidium Bromide

RNA EZ-Vision® versus Ethidium bromide staining

Total RNA was purified from K562 cells with Ribozol™ RNA Extraction Reagent. Samples were denatured at 65°C for 10 minutes in RNA EZ-Vision® loading buffer containing either 0.003% ethidium bromide or RNA EZ-Vision® dye. The denatured RNA was loaded onto a 2% formaldehyde denaturing agarose gel (Agarose I™) and resolved for 1.5 hours at 5.1 V/cm. Bands were visualized by UV illumination and image capture was performed with a Syngene GBox-HR Gel Doc System with a SYBR® Green filter.

This 4X loading buffer solution includes fluorescent protein dye that comigrates with the protein-SDS complex during electrophoresis. Solution can be stored at room temperature. Eliminates the need for post-staining and destaining and is compatible with downstream blotting applications.

Immediate Visualization
Instant visualization of protein bands upon UV illumination of SDS-PAGE gels. Sensitivity of detection goes down to 100ng protein.

Sensitivity of Protein EZ-Vision vs. Coomassie

Sensitivity of Protein EZ-Vision®

Protein EZ-Vision® (A) compared to Coomassie® Blue sensitivity (B). Both figures were generated using the same gel for comparison. Two-fold serial dilutions of bovine serum albumin (BSA) were separated via a 12.5% Laemmli SDS-PAGE gel using Protein EZ-Vision®, 4X as the sample loading buffer. Lane 7 is approximately 100ng protein. Immediate fluorescent visualization of protein post-electrophoresis was achieved with a 2 sec UV exposure using a SYBR® Green filter with the Syngene GBox-HR Gel Doc System (A). The same gel was post-stained with Coomassie® Blue protein stain for one hour and destained one hour (B).
