High-Performance Liquid Chromatography, or HPLC, is a type of chromatography commonly used to characterize and purify proteins or peptides from complex mixtures of molecules. Reversed-Phase HPLC is a form of the technique most widely used, which utilizes a hydrophobic column as the stationary phase and a non-polar solvent as a mobile phase. Elution of solutes from the column is dependent on the hydrophobicity of the solvents and is achieved by addition of organic solvent to the mobile phase. Commonly used solvents include methanol and acetonitrile with water. An ionic modifier, such as trifluoroacetic acid, may be included in the solvents.
