RapidBlock™ Solution, 10X

AMRESCO’s RapidBlock™ Solution reduces blocking time to 5 minutes for Western and dot blotting procedures on PVDF and nitrocellulose membranes. This protein-free formulation minimizes cross-reactivity and non-specific antibody binding, generating blots with low background and enhanced signal-to-noise ratio. Results with RapidBlock™ meet or exceed those obtained with buffers containing dried milk or BSA, which both require 1 hour of blocking time.

Time course of blocking by RapidBlock™

Cytoplasmic proteins from K562 cells were resolved on a 12.5% Fluorescent SPRINT NEXT GEL® (M318) and then transferred to a PVDF membrane.  The membrane was cut into strips and blocked 5 or 30 minutes in either TBST/5% non-fat dry milk (lanes 1,2) or 1X RapidBlock™ Solution (lanes 3,4).  The strips were probed with anti-β-tubulin primary antibody and a HRP-conjugated secondary antibody, followed by detection with VisiGlo Plus™ HRP Chemiluminescent Substrate Kit (N219).  Results demonstrate that 5 minute blocking in RapidBlock™ Solution is sufficient to achieve low background and an enhanced chemiluminescent signal compared to blocking for equivalent time in TBST/5% non-fat dry milk.

Chemiluminescent signal enhancement with RapidBlock™

A 12.5% Fluorescent SPRINT NEXT GEL™ (M318) was loaded with 10µg per lane of cytoplasmic protein isolated from K562 cells using AMRESCO's Cytoplasmic/Nuclear Protein Enrichment Kit (M330) (Figure A).  Proteins were resolved at 300 volts for 30 minutes and then visualized on a UV transilluminator (figure A) prior to transfer to a PVDF membrane.  The membrane was cut into strips and blocked in either TBST/5% non-fat dry milk (figure B, lane 1) or RapidBlock™ Solution (figure B, lane 2).  The blots were probed with anti-β-tubulin primary antibody and then HRP-conjugated secondary antibody, followed by detection with VisiGlo Plus™ HRP Chemiluminescent Substrate Kit (N219).
