LP-NEXT GEL® KIT is a revolutionary product for the resolution of large proteins on electrophoretic gels. The kit employs an agarose matrix and a customized SDS running buffer to resolve denatured polypeptides ranging from 14 kDa to greater than 6,000 kDa. Gels are run on conventional horizontal electrophoresis equipment including mini-submarine apparatuses. LP-NEXT GEL® is recommended for studies of a variety of large polypeptides including cytoskeletal and muscle proteins, nuclear scaffolding proteins, cross-linked molecules and other proteins too large to clearly resolve on conventional SDS-PAGE. LP-NEXT GEL® is compatible with most downstream applications such as Western Blotting, N-terminal sequencing and MALDI.

An optional fluorescent dye that allows nearly instantaneous band visualization and eliminates the need for post-run staining and destaining is included in the kit. The fluorescent dye is included in the gel matrix and binds to the migrating proteins during electrophoresis. After the run, the protein-dye complex fluoresces after a brief exposure (less than 5 minutes) to UV light. Background fluorescence is virtually absent since the unbound probe does not emit light upon irradiation. Sensitivity of the fluorescent stain is comparable to Coomassie® Blue. Fluorescent LP-NEXT GEL® is ideal for applications where immediate visualization of protein bands is desired. The fluorescent dye is compatible with Western Blotting. The effect of the fluorescent reagent on MALDI and sequencing procedures is under investigation.

Each LP-NEXT GEL® KIT contains Agarose HRP, Fluorescent NEXT GEL® Buffer, 20X, NEXT GEL® Running Buffer, 20X and NEXT GEL® Sample Loading Buffer, 4X.
